A team of German researchers has revealed the potential to enhance the effectiveness of the traditional drug “Tamoxifen” by combining it with a supplemental compound known as “Z-Endoxifen.” This breakthrough may represent significant medical progress for women who do not respond well to current treatments.

Tamoxifen is one of the leading medications used to treat hormone-receptor-positive breast cancer, working by blocking the binding of the hormone estrogen to cancer cells, thus limiting tumor growth. However, its effectiveness decreases in about one-third of patients due to reduced levels of the enzyme “CYP2D6,” which is responsible for converting the drug into its active form in the body.

The research, published in the journal “Clinical Cancer Research,” indicated that supplying patients with “Z-Endoxifen” as a supplement helps overcome this biochemical hurdle and enhances treatment efficacy in those who experience poor metabolic conversion.

The study involved 235 patients in the early stages of the disease, with one group receiving Tamoxifen alone while another received the combination treatment with Z-Endoxifen. Results showed that active drug levels in the blood were similar across both groups, with only minor and comparable side effects recorded.

Matthias Schwab, leader of the research team at the Margaret Fischer-Bosch Institute in Stuttgart, stated that these results represent “the first solution to a long-standing problem of many patients’ poor response to Tamoxifen.” He added that the new approach “could pave the way for more precise and effective treatment protocols in the future.”

Researchers hope this discovery will lead to the development of tailored treatments considering genetic differences among patients, thereby improving cure rates and reducing the side effects of traditional therapy.
